  • To test the new dbem1dbem3 +pGal strain in different Gal concentrations.

14.3. Method#

  • 1ul of glycerol stock in 100ul media

  • incubation in 30C in the biotek

  • Media base: 4xCSM-NF+2%Raff

  • Plate layout:

14.4. Results#

  • End of incubation , after 48 hours

  • Dilution and start measuring

    • 1ul of incubated culture into 100ul fresh media

    • 36C protocol

  • ylic140a/b did not grow in 0% Gal !!!

14.4.1. 2nd experiment to test the measuring temperature#

  • Change to 30C during measurement.

  • Incubation : 09082021 (same layout as the previous experiment) End of incubation

  • I suspect that well B10 and B11 are contaminated…..

14.5. Conclusion#

  • It seems at 30C the effect of bem1d is stronger than at 36C.
